Abstract

We established modified plastic embedding techniques for undecalcified bone marrow
core biopsy specimen, using Historesin and Immuno- BedTM. To save the
embedding medium, a wooden holder was devised, which fit with Histomold
TM(LKB, Sweden) concave portion.
All the procedures from fixation to routine staining took 36-48 hours, that was equal
length of time to that of paraffin embedding method.
Sections were cut at 3¥ìm with tungsten carbide knife and were attached to slides by
warming on the slide warmer at 100¡É for 4 minutes.
May-Grunwald Giemsa(MGG) and H&E stains were done with modified procedures.
The plastic embedding methods produced thin sections of undecalcified osseous
specimens. The thin sections had significant advantages over paraffin sections in
identification of individual cells and recognition of the structural alterations, that have
broadened the understanding of bone marrow structures.
